Introduction: The COVID-19 pandemic may have heightened stress levels among university professors, exacerbating workplace challenges. Objective: To analyze the prevalence and the demographic, lifestyle, work-related, and mental health factors associated with high stress levels among university professors during the COVID-19 pandemic. Method: An observational, cross-sectional, and quantitative study was conducted with 607 professors from public institutions in Mato Grosso. Descriptive and inferential analyses were performed. Results: A total of 51.56% of professors reported high levels of work-related stress. In the final model, the following variables remained significantly associated: the impact of work on mental health (p<0.001), lack of physical and material resources (p=0.001), dissatisfaction in relationships with colleagues (p=0.022), lack of professional engagement (p=0.022), depression (p<0.001), and sleep disorders (p=0.002). A temporary employment contract (p=0.023) was identified as a protective factor. Discussion: The high stress levels reported may have adverse effects on both health and teaching quality. Conclusion: These findings provide a situational overview that can inform policies and strategies aimed at promoting well-being in the university environment at the national level.
